Application of statistical experimental methods to optimize medium for exopolymer production by newly isolated Halobacterium sp. SM5 Electron. J. Biotechnol.

The study investigated on medium optimization for production of exopolymer by a newly isolated bacterium, Halobacterium sp. SM5, using the mixture design and response surface method. The mixture experiment was designed by setting five parameters at the lowest and highest of concentrations: pseudo components design containing 0.15-0.25 glucose, 0.15-0.25 yeast extract, 0.35-0.45 MgSO4·7H2O, 0.1-0.2 vitamin casamino acid and 0.02-0.06 KCl. The results of the mixture design revealed that the effect of nutrients or elements on the exopolymer produced by the strain SM5 were in the order of MgSO4·7H2O > yeast extract > vitamin casamino acid > KCl > glucose, respectively. High-temperature ethanol production using thermotolerant yeast newly isolated from Greater Mekong Subregion BJM

Abstract The application of high-potential thermotolerant yeasts is a key factor for successful ethanol production at high temperatures. Two hundred and thirty-four yeast isolates from Greater Mekong Subregion (GMS) countries, i.e., Thailand, The Lao People's Democratic Republic (Lao PDR) and Vietnam were obtained. Five thermotolerant yeasts, designated Saccharomyces cerevisiae KKU-VN8, KKU-VN20, and KKU-VN27, Pichia kudriavzevii KKU-TH33 and P. kudriavzevii KKU-TH43, demonstrated high temperature and ethanol tolerance levels up to 45 °C and 13% (v/v), respectively. Optimization of culture conditions for exopolysaccharides production in Rhizobium sp. using the response surface method Electron. J. Biotechnol.

The combined effects of the processing parameters for exopolysaccharides production by Rhizobium sp. was studied using the experimental design and response surface methodology. The experiments were carried out using a fermenter with 20 L capacity, as the reactor. All processing parameters were online monitored. The temperature [(30 ± 1)ºC] and pH value (7.0 ± 0.1) were kept constant throughout the experimental time. Optimization of nutrition factors on chitinase production from a newly isolated Chitiolyticbacter meiyuanensis SYBC-H1 BJM

The present study reports statistical medial optimization for chitinase production by a novel bacterial strain isolated from soil recently, which the name Chitinolyticbacter meiyuanensis SYBC-H1 is proposed. A sequential statistical methodology comprising of Plackett-Burman and response surface methodology (RSM) was applied to enhance the fermentative production of chitinase, in which inulin was firstly used as an effective carbon source. As a result, maximum chitinase activity of 5.17 U/mL was obtained in the optimized medium, which was 15.5-fold higher than that in the basal medium. The triplicate verification experiments were performed under the optimized nutrients levels which indicated that it well agreed with the predicted value.
